WG 5 – Objectives

This WG focuses on the microscope lateral and axial resolution, which is essential for reporting size measurements of near- resolution limit objects or distances between them. Resolution is highly related to the objective quality but depends strongly upon other parameters ranging from the sample preparation to the signal detection.

The WG aims to define sample preparation, image acquisition, and data analysis protocols for testing resolution, first using sub-resolution fluorescent bead preparations and second employing alternative pattern-based methods. Monitoring the resolution (Point Spread Function in the case of beads) over time will identify possible aberrations in the system. Pooling the data from multiple laboratories within the WG will allow them to compare reproducibility for sample preparation, data acquisition, and data analysis tools, thereby determining a robust, easy-to-use protocol to propose to the community.

Meeting summary

The protocol V2 was discussed and it was voted to pass the next version of the protocol to the SC. For the upcoming ELMI workshop, plans were initiated to host a one-hour session, considering point source and line patterns, and coordinating tasks among participants. An update on the paper shared insights on confocal SBR and r² values, noting differences due to noise factors. Discussions included data analysis techniques and the challenges of data variance over time. The meeting concluded with a call for organizing data in OMERO and an update on a new ISO norm for metadata, inviting community participation. The next meeting is postponed by a week to accommodate a French national holiday.

Meeting summary

We discussed the ELMI workshop, including how it went and what we could improve. We also covered the process of OMERO uploads and linking them to Airtable for tracking results, as well as the filenaming nomenclature.
